Effect of NH4+, Ca++ and Mg++ Ion Concentrations on Organogenesis and Nutrient Content through Cymbidium Protocorm Culture


Abstract
This study was carried out to investigate the effects of NH©þ^+, Ca^(++) and Mg^(++) ions which could have influences on the protocorm proliferation and organogenesis through protocorm culture of Cymbidium elcapitan `Conquestador¢¥ obtained by shoot tip culture. The relations between inorganic nutrient in the medium and those in the plant tissue were also studied.
Best shoot formation and growth occurred in a medium with 4 mM of NH©þ-N while good root formation and growth were obtained from no NH©þ-N treatment. As the NH©þ-N level in the medium increased, potassium content in the tissue decreased, whereas P and Mg contents increased slightly.
Eight mM of calcium in the medium promoted formation and growth of shoots but 4 mM enhanced those of roots. Increasing Ca levels in the medium tended to decrease the chlorophyll content, N, K and Mg contents in the tissue.
Magnesium ion concentration of I mM promoted shoot formation and growth, but root formation and growth were promoted on the medium containing no Mg. The tissues grown on the medium without Mg ion had prominently low concentrations of N and Ca content. But as the ion level of Mg increased, Ca content decreased in all the tissue analyzed.
